TRKB Effector Reporter Cell
| I. Background | |
| TRKB(原肌球蛋白受體激酶 B,亦稱 BDNF 受體或 NTRK2)是神經(jīng)營養(yǎng)因子受體家 族(Trk 受體)的關(guān)鍵成員。作為一種跨膜激酶受體,其特異性結(jié)合配體腦源性神 經(jīng)營養(yǎng)因子(BDNF) 及神經(jīng)營養(yǎng)因子-4(NT-4),通過激活下游 PI3K/Akt、Ras/MAPK 和 PLCγ信號通路,核心調(diào)控神經(jīng)元存活、突觸可塑性、神經(jīng)遞質(zhì)釋放及軸突導向,是維持 學習記憶、情緒平衡與神經(jīng)發(fā)育的核心樞紐。在病理狀態(tài)下,TRKB 信號功能障礙直接參 與抑郁癥、阿爾茨海默病、理性疼痛及癲癇等神經(jīng)系統(tǒng)疾病,同時作為泛癌靶點, 其基因融合事件可驅(qū)動肉瘤、甲狀腺癌等實體瘤進展(拉羅替尼/恩曲替尼等 TRK 抑制劑 已獲批治療),而代謝領(lǐng)域則涉及肥胖(下丘腦 TRKB 調(diào)控能量平衡)與非酒精性脂肪肝 (肝星狀細胞 TRKB 促纖維化),目前針對 TRKB 的靶向策略涵蓋小分子激酶抑制劑(抗 癌)、BDNF 模擬肽(神經(jīng)保護)及基因療法(遞送 BDNF),但需克服血腦屏障穿透、 組織選擇性及耐藥突變(如 TRKB G667C)等挑戰(zhàn)。 | |
